The objective of the present work was to evaluate the influence of flow cytometry analysis and cell sorting (FACS) conditions on the detection of Cryptosporidium oocysts and Giardia cysts in surface water concentrates. FACS has been used to discriminate and separate the parasites from most of the contaminated particles and therefore facilitate microscopic observation, both in labor and in reliability of the assessment of the typical characteristics of the parasites. FACS also allowed larger volumes of water concentrates to be analyzed, thus increasing the sensitivity of the method. This study was done in two laboratories using two different FACS conditions and settings. The FACS discriminated the particles based on their green fluorescence intensity versus their Forward Scatter (size) or their Side Scatter signal (granularity). The two laboratories each produced concentrates by several procedures (polypropylene cartridge filter, compressed foam disc filters and polyethersulfone capsule filter and ferric sulphate flocculation) from spiked water samples. The concentrates were processed with and without flotation and examined by microscope, with and without use of flow cytometry and cell sorting. Includes 11 references, tables, figures.
