This powerpoint presentation begins by stating the study motivation that includes the following: Current Regulatory Framework - protection of public health from pathogens such asCryptosporidium (LT2), andmanagement of DBPs and organics (Stage 2 D/DBP Rule); and,Chlorine Disinfection- effective for many microorganisms under properconditions,- some viruses (adenovirus) and protozoa (Cryptosporidium)resistant to chlorination, and- reacts with organic matter to form regulated DBPs. A brief overview of sonication is presented, along with a flow-through cavitation unit. Experimental objective: quantify E. coli inactivation, known concentration of organisms in chlorine demand free PBS, disinfection by sonication, chlorination, or both, and monitored E. concentration, power output, and temperature. Experimental methods included the following: E. coli Culturing and Enumeration- E. coli cultured in laboratory to stationary phase, centrifuged and decanted,- resuspended in phosphate buffered saline (PBS) at startingconcentration of ~107 cells/mL,- enumerated by pour plates on tryptic soy agar. Preliminary results and related research are provided. Includes table, figure.
